Reversed-phase chromatography is the preferred HPLC technique for the analysis of many drug substances, but many alternative C18 columns show strong adsorption of basic drugs

Agilent Technologies Europe has announced rapid, reliable and precise analyses of antibiotic, antidepressant, and antibacterial drugs using high-performance liquid chromatography (HPLC) and Agilent Zorbax SB-C18, Eclipse XDB-C18, and SB-Aq HPLC columns.

Reversed-phase chromatography is the preferred HPLC technique for the analysis of many drug substances.

In this technique, many C18 columns in the industry show strong adsorption of basic drugs, frequently leading to peak tailing, fronting, and poor resolution.

Zorbax HPLC columns are designed to reduce or eliminate adsorption when used for drug testing with water-organic mobile phases.

It is well known that highly polar drugs are poorly retained on common C18 columns and often show poor resolution.

Furthermore, these long chain alkyl phases can undergo phase collapse when used with highly aqueous mobile phases.

Phase collapse results in poor peak shape, nonreproducible retention times, and slow equilibration after gradient elution.

The Zorbax SB-Aq HPLC column was designed to eliminate phase collapse and still retain hydrophilic drug substances when exposed to highly aqueous mobile phases.

Agilent chemists have demonstrated that in all cases, the Zorbax columns achieved excellent peak shape, good selectivity, rapid resolution, excellent stability at neutral or low pH, and low-level detection of subject drugs at low buffer concentration.
