Pichia pastoris is sensitive to concentrations of methanol which need to be maintained within a narrow range.
As well as being necessary to control induction and regulation of the promoter for the desired gene expression, methanol also serves as a carbon source during the growth phase.
However, high levels of methanol are cytotoxic and the accumulation of the metabolite formaldehyde must also be controlled.
With quick and accurate measurements available using the YSI 7100, methanol levels can be monitored to ensure they remain at optimum levels for high protein yield without becoming cytotoxic.
