Resequencing system for mutational
profiling

Applied Biosystems has launched the VariantSEQr resequencing system, said to be the first complete, cost-effective solution for mutational profiling The VariantSEQr resequencing system comprises high-quality ready-to-use primers, pre-mixed PCR reagents, BigDye Terminator v3.1 sequencing reagents and SeqScape software v2, complete with simple robust PCR and sequencing protocols to streamline the whole laboratory process

The system has been under evaluation at the Genome Sequencing Center, Washington University School of Medicine, and the centre's co-director, Elaine Mardis, explained: "When we began to compare mutation profiles in tumours from acute myelogenous leukaemia patients, we quickly realised just how tedious and expensive it is to select, design and test PCR primers.
"The VariantSEQr resequencing system has eliminated these time-consuming and costly steps and obtains significantly higher PCR first time pass rates than we can achieve with our own primer sets".
Bob Welch, deputy director of the core genotyping facility in the centre, added: "In two months the lab was able to resequence seven candidate cancer gene coding regions, comprising 152 amplicons in 102 individuals.
"Analysis of the data generated by the study showed 133 known SNPs but, more importantly, identified 170 novel SNPs.
"Using the VariantSEQr resequencing system we were able to reduce project time by more than a month and the number of PCR reactions required by almost half".
The VariantSEQr resequencing system is available for use on the Applied Biosystems 3730xl and 3730 DNA analysrs or the ABI Prism 3100 and 3100-Avant genetic analysers, and can be ordered through the myScience webpage.
Applied Biosystems says it expects to release additional genes based on feedback from researchers.
