Columns for TFA-free LC/MS

PepMap columns from Dionex are designed for the separation of peptide mixtures, and require only one-tenth of the TFA concentration in the mobile phase

PepMap, a silica-based stationary phase where virtually all silanophobic activities are suppressed, is available on columns with an ID ranging from 50æm to 1.0mm The use of the PepMap material allows the separation of peptides with a minimal bandbroadening

One of the main advantages of using PepMap is the fact that only one-tenth the concentration of TFA in the mobile phase is required.

As a rule, TFA concentration needs to be only as high as 0.005% to achieve symmetric peaks.

With PepMap, it is even possible to exchange the TFA with aspetic acid, 0.05%.

In both cases an increased signal/noise ratio is achieved, especially when using an ESI-MS detector.

Due to these characteristics, PepMap is suited for separation of peptide mixtures, obtained by, for instance, tryptic digestion of proteins.

This is even more relevant when using a mass spectrometer as detector.
