Routine analysis of oxidative DNA damage
HPLC method using multi-channel electrochemical array detection plus automated column switching procedure to measure 8-hydroxy-2'-deoxyguanosine (8OH2'dG) in urine
ESA Laboratory Services has developed a highly specific, sensitive, interference-free, and accurate HPLC method using proprietary multi-channel electrochemical array detection technology in conjunction with an automated column switching procedure to measure 8-hydroxy-2'-deoxyguanosine (8OH2'dG) in urine - a prime indicator of oxidative DNA damage.
8-hydroxy-guanine, a highly mutagenic DNA adduct, can be formed by exposure of DNA to a number of oxidising species, such as hydroxyl free radicals.
In vivo repair mechanisms remove this adduct as 8OH2'dG, which is excreted in the urine.
Urinary 8OH2'dG is currently being used as an indicator of oxidative stress for ageing and various diseases such as Alzheimers, diabetes, and cancer.
ESA's fully validated HPLC method is the only commercially available assay measuring the extremely low levels of 8OH2'dG in urine, a very complex matrix.
Additionally, analyses of 8OH2'dG in plasma, saliva, sweat or CSF are available.
In extracted, hydrolysed nuclear or mitochondrial DNA, the analysis is extended to include the precursor 2'dG.
