Optimising column-based DNA extraction

Integra Biosciences has announced a new application note describing the use of the Vacusafe Comfort for optimising DNA purification based upon the widely used bind-wash-elute procedure This involves releasing the nucleic acids from the cells, binding the DNA onto a matrix (usually packed in a column), washing impurities off the matrix-bound DNA and finally eluting the DNA

In the described experiment plasmid DNA (pBluescript) from an overnight culture of transformed DH5a E coli was extracted with a commonly used kit.

To investigate the effect of different levels of applied vacuum on the effectiveness of the DNA purification, the cell lysate containing the DNA was drawn through a matrix-packed column using a range of applied vacuum.

The results of the experiment clearly showed that the DNA extraction kit performed best at lowest vacuum settings, probably because this allowed maximal binding of DNA onto the column matrix.
