Assay allows monitoring of gene
downregulation
Luciferase assay system allows quantitation of two reporter gene outputs from a single sample with just one reagent addition
Quantitation of two reporter gene outputs in a single sample is now possible with Promega's Chroma-Glo luciferase assay system Furthermore, both red and green luminescent signals are produced following the addition of a single reagent meaning the assay is readily adaptable to high throughput applications especially where sample well size is limiting

The assay is very easy to use: simply add a single volume of the Chroma-Glo reagent directly to cells in growth medium and then measure the signal in a dual filter luminometer.
The signal is very stable (half-life in excess of 30 minutes), allowing batch processing of multiple plates without multi-step handling.
Promega has engineered red- and green-emitting luciferases that are almost identical.
At the genetic level this minimises potential RNA and protein effects on reporter expression and hence enables simple and effective normalisation of an experimental reporter to a control reporter.
The monitoring of downregulation of genes has proved difficult but the Chroma-Glo assay simplifies the process.
The end result is decreased inter- and intra-sample variability.
Alternatively, both reporters can be used to multiplex experimental measurements, increasing the amount of information obtained from a single assay.
