Promega, supplier of products for RNAi (ddRNAi) technologies in non-therapeutic applications, has expanded its range of RNAi tools with two new products; psiCHECKT-1 and psiCHECKT-2 vectors, and the siSTRIKET U6 Hairpin Cloning System psiCHECKT-1 and psiCHECKT-2 vectors provide an effective means to validate the results of RNAi experiments and provide a rapid and quantitative approach for the initial optimisation of RNAi

These unique vectors use luminescence based reporter technology to directly quantify the siRNA silencing effect and to screen out phenotypic artefacts caused by transfection protocols or assay conditions.
psiCHECKT-1 is designed for use with living cells, whereas psiCHECKT-2 is suitable for end point lytic assays.
The siSTRIKET U6 Hairpin Cloning System consists of a series of ready-to-use linearised plasmids designed for fast and easy cloning of hairpin target sequences for the expression of siRNA in human cells.
The Systems offer a choice of eukaryotic antibiotic-selectable resistance markers, allowing stable transfection for long term studies of silencing effects over a period of several weeks, rather than the 72 hours common to synthetic RNA based approaches.
In addition, the antibiotic markers ensure that the entire cell population expresses the siRNA construct, thereby simplifying phenotypic analysis and solving problems of low transfection efficiencies.
Also in the RNAi Research Range are: The T7 RiboMAXT Express RNAi System - an in vitro transcription system designed for the rapid, protein-free production of milligram amounts of double-stranded RNA suitable for mammalian and non-mammalian cells.
The dsRNA's produced in this system have been shown to produce silencing effects equivalent to chemically synthesised synthetic siRNA.
And the CodeBreakerT siRNA Transfection Reagent - a ready-to-use reagent optimised for efficient transfection of siRNA and enabling effective gene silencing in a variety of eukaryotic cell lines with minimal cell death.
