Halochip gives protein interactions that provide greater sensitivity than chromatin immunoprecipitation techniques without the need for a specific antibody and in half the time

Halochip from Promega replaces antibody-based chromatin immunoprecipitation methods Promega announce the release of Halochip, a novel approach to studying DNA

Removing the need for a specific antibody means the Halochip system is far more flexible and much faster than existing methods.

The system's tolerance to stringent washing means background signals are reduced allowing the detection of small changes in protein binding patterns using minimal numbers of cells.

Proteins of interest are expressed as Halotag fusion proteins and cross-linked to their DNA binding sites with formaldehyde.

The Halotag protein allows immobilisation of this DNA:Protein complex to Halolink resin while the sample is washed to remove non-specific DNA and proteins.

The cross-linking is then reversed to release the target DNA for analysis.

The introduction of the Halochip system is a further addition to the range of assays that are facilitated using Promega's Halotag technology.

This unique fusion protein system allows a single construct to be used for multiple applications, including imaging, immobilisation, protein:protein interactions and protein purification.
